Monitoring the fate of epigenetic cues delivered by sperm to the embryo can be challenging due to the paucity of available material. The use of Xenopus laevis egg extract to mimic the chromatin events associated with the first embryonic division provide an alternative approach to the use of embryos. In this chapter, we describe a protocol to carry out quantitative ChIP-seq analysis of modified histones fate following sperm exposure to maternal factor components to understand the relevance of male-derived histone modifications during embryogenesis.
